For an Uchiha, the formidable clan his father fought on a regular basis, this man didn't really fit. He seemed... kind of weak, actually. And the way his eyes moved.
"You can't see us, can you?" he asked, not really expecting the man to answer. After all, what shinobi reveals that kind of weakness.
But Itachi knew who Hashirama was. He knew this was the future first Hokage. And he also knew, if he was going to get anywhere, he had to trust them both and vice versa.
